Indications
- Palliative treatment of advanced carcinoma of the breast or endometrium
- Treatment of anorexia, cachexia, or unexplained weight loss in AIDS & cancer patients
Pharmacology
- Synthetic, antineoplastic and progestational drug
- Inhibition of pituitary gonadotrophin production
- Decrease in estrogen secretion
- Modifying action of other steroid hormones
- Cytotoxic effect on tumor cells
- Estrogen receptor mechanism
Dosage & Administration
- Tablet dosage for breast cancer: 160 mg/day
- Tablet dosage for endometrial carcinoma: 40-320 mg/day in divided doses
- Oral suspension dosage: 800 mg/day (20 ml/day)
Interaction
- No investigated interactions with concomitant medications
- Possible fluid retention with high dosage or prolonged use
Contraindications
- Hypersensitivity to Megestrol Acetate
- Known or suspected pregnancy
Side Effects
- Weight gain (associated with increased appetite)
- Thromboembolic phenomena including thrombophlebitis and pulmonary embolism
- Glucocorticoid effects
- Nausea
- Dyspnea
- Hyperglycemia
- Hypertension
- Alopecia
- Mood changes
Pregnancy & Lactation
- Pregnancy Category D
- Excretion in mother's milk (approximately 0.1%)
- Discontinuation of nursing during treatment with Megestrol Acetate
Precautions & Warnings
- Close surveillance for patients with recurrent or metastatic cancer
- Use with caution in patients with a history of thromboembolic disease
- Exacerbation of preexisting diabetes
Use in Special Populations
- Safety and effectiveness in pediatric patients not established
- Insufficient data for patients 65 years of age and older
Overdose Effects
- No serious unexpected side effects from studies involving high dosages
