Indications
- Endometriosis
- Benign breast disease

- Preoperative thinning of the endometrium before hysteroscopic endometrial ablation
Pharmacology
- Suppression of pituitary-ovarian axis
- Depression of preovulatory surge in FSH and LH
- Inhibition of ovarian steroidogenesis
- Binding to androgen, progesterone, and glucocorticoid receptors
- Increase in metabolic clearance rate of progesterone
- Suppression of ovarian function
- Correcting biochemical deficiency in hereditary angioedema
Dosage & Administration
- Usually given in up to 4 divided doses
- Treatment should start during menstruation in women of child-bearing potential
- 200-800 mg daily for endometriosis
- 100-200 mg daily for menorrhagia
- 100-400 mg daily for severe cyclical mastalgia
- 300 mg daily for benign breast cysts
- 400 mg daily for gynaecomastia
- 400 mg-800 mg daily for pre-operative thinning of endometrium
Interaction
- Increased insulin resistance with insulin
- Potential interaction with exogenous estrogens and/or progestogens
- Effect on plasma concentration of anticonvulsants, antihypertensives, cyclosporine, and migraine therapy
Contraindications
- Pregnancy and breast-feeding
- Impaired hepatic, renal, or cardiac function
- Porphyria
- Thromboembolic disease
- Androgen-dependent tumor
- Abnormal vaginal bleeding
- Hypersensitivity to danazol
