Pharmacology

Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). It does not bind directly to GABA receptors but binds with high affinity to the alpha2-delta site in central nervous system tissues, which may be involved in its anti-nociceptive and antiseizure effects.

Dosage

DPN: Maximum recommended dose of Pregabalin is 100 mg three times a day for patients with creatinine clearance of at least 60 ml/min
PHN: Recommended dose of Pregabalin is 75 to 150 mg two times a day or 50 to 100 mg three times a day
Fibromyalgia: Recommended dose of Pregabalin is 300 to 450 mg/day
Spinal cord injury: Recommended dose range of Pregabalin is 150 to 600 mg/day
Special Instructions: Pregabalin CR tablet should be administered after an evening meal and swallowed whole

Route: Oral
Instructions: Can be taken with or without food. Pregabalin CR tablet should be administered after an evening meal and swallowed whole.

Interaction

Drug: Unlikely to be involved in significant pharmacokinetic drug interactions

Contraindications

Pregabalin is contraindicated in patients with known hypersensitivity to Pregabalin or any of its components

Pregnancy & Lactation

Pregnancy: No adequate and well-controlled studies; potential risk to fetus
Lactation: Small amounts detected in the milk of lactating women; breastfeeding not recommended

Precautions & Warnings

Angioedema can occur and may be associated with life threatening respiratory compromise requiring emergency treatment
Discontinue immediately in case of hypersensitivity reactions
Increase in risk of suicidal thoughts or behavior
Respiratory depression may occur when used with concomitant CNS depressants
May cause dizziness and somnolence, impairing ability to drive or operate machinery
Increased seizure frequency or other adverse reactions if rapidly discontinued
May cause peripheral edema
Caution should be exercised when coadministering with thiazolidinedione antidiabetic agents

Use in Special Populations

Children and Adolescents: Safety and effectiveness not established for various uses

Overdose Effects

Symptoms include reduced consciousness, depression/anxiety, confusional state, and seizures; no specific antidote
