Indications
- Palliative treatment of advanced carcinoma of the breast
- Palliative treatment of advanced carcinoma of the endometrium
- Treatment of anorexia in patients with a diagnosis of Acquired Immunodeficiency Syndrome
- Treatment of cachexia in patients with a diagnosis of cancer
- Treatment of unexplained, significant weight loss in patients with a diagnosis of Acquired Immunodeficiency Syndrome
- Treatment of unexplained, significant weight loss in patients with a diagnosis of cancer
Pharmacology
- Synthetic, antineoplastic and progestational drug
- Inhibition of pituitary gonadotrophin production
- Resultant decrease in estrogen secretion
- Modifying the action of other steroid hormones
- Direct cytotoxic effect on tumor cells
- Estrogen receptor mechanism
- Estimates of plasma levels of Megestrol Acetate
Dosage & Administration
- Dosage for breast cancer treatment: 160 mg/day
- Dosage for endometrial carcinoma treatment: 40-320 mg/day in divided doses
- At least 2 months of continuous treatment for efficacy determination
- Recommended adult initial dosage of Megestrol Oral Suspension: 800 mg/day (20 ml/day)
Interaction
- Possible interactions of Meget with concomitant medications
- Estrogens and progestogens may cause fluid retention
Contraindications
- History of hypersensitivity to Megestrol Acetate
- Known or suspected pregnancy

- Thromboembolic phenomena including thrombophlebitis and pulmonary embolism

- Tumor flare
- Hyperglycemia
- Glucose intolerance

- Mood changes
- Hot flashes
- Malaise
- Asthenia
- Lethargy

- Rash
Pregnancy & Lactation
- Pregnancy Category D
- Not recommended during the first four months of pregnancy
- Small amounts excreted in mother's milk
- Potential adverse effects on the newborn
- Nursing should be discontinued during treatment
Precautions & Warnings
- Close surveillance for patients treated for recurrent or metastatic cancer
- Use with caution in patients with a history of thromboembolic disease
- Exacerbation of preexisting diabetes with increased insulin requirements
Use in Special Populations
- Safety and effectiveness in pediatric patients have not been established
- Insufficient data from clinical studies for patients 65 years of age and older
Overdose Effects
- No serious unexpected side effects from studies involving Meget administered in dosages as high as 1600 mg/day
