Indications
- Monotherapy for partial-onset seizures in adults and adolescents
- Adjunctive therapy for partial-onset seizures in adults, adolescents, children, and infants
- Treatment of myoclonic seizures in adults and adolescents
- Treatment of primary generalized tonic-clonic seizures in adults and adolescents
Description
Mechanism of action is unknown, inhibits burst firing without affecting normal neuronal excitability
Dosage
- Recommended starting dose for adults and adolescents is 250 mg twice daily
- Maximum dose is 1500 mg twice daily for adults and adolescents
- Tablet formulation not adapted for infants and children under 6 years
Administration
Must be taken orally, swallowed with sufficient quantity of liquid, and may be taken with or without food
Interaction
- No influence on serum concentrations of existing antiepileptic medicinal products
- Inhibited renal clearance of the primary metabolite with probenecid
- Decreased methotrexate clearance when administered concomitantly
Contraindications
Hypersensitivity to the active substance or other pyrrolidone derivatives or excipients
Side Effects
- Most common adverse reactions include somnolence, asthenia, infection, and dizziness in adults
- Fatigue, aggression, nasal congestion, decreased appetite, and irritability in pediatric patients
Pregnancy & Lactation
Not recommended during pregnancy and breastfeeding, excreted in human breast milk
Precautions & Warnings
- Dose adjustment required for patients with renal impairment
- Rare cases of decreased blood cell counts described at the beginning of treatment
Use in Special Populations
Dose adjustment recommended for elderly patients and those with compromised renal function
Overdose Effects
May lead to somnolence, agitation, respiratory depression, and coma
