Indications
- Bronchial asthma
- Bronchospasm
- Chronic obstructive pulmonary disease (COPD)
- Pulmonary disease with spastic bronchial component
Pharmacology
Doxophylline selectively inhibits phosphodiesterase 4, relaxes bronchial smooth muscle, inhibits platelet activating factor (PAF) and generation of leukotriene production
Dosage & Administration
- Elderly: 200 mg tablet two or three times daily
- Adults: 400 mg tablet two or three times daily
- Children:
- >12 years: 10 ml syrup or 200 mg tablet two or three times daily
- 6-12 years: 6-9 mg/kg body weight two times daily
Interaction
- Interactions: Doxobron should not be administered with other xanthine preparations, limit consumption of caffeine-containing beverages and food, caution in administering with ephedrine or other sympathomimetic drugs
- Drugs that may affect:
- Erythromycin
- TAO
- Lincomycin
- Clindamycin
- Allopurinol
- Cimetidine
- Influenza vaccine
- Propanolol
- Phenytoin
- Other anticonvulsants
- Cigarette smoking
Contraindications
Known hypersensitivity to the drug or other xanthine derivatives, acute myocardial infarction, hypotension, lactation

- Extrasystoles
- Tachypnea
- Hyperglycemia
- Albuminuria
Pregnancy & Lactation
- Use during pregnancy should be evaluated carefully
- Contraindicated during lactation
- No sufficient clinical evidence about effects during pregnancy
- No interference with ability to drive and use machines
Precautions & Warnings
- Factors affecting hepatic clearance: Age, congestive cardiac decompensation, chronic obstructive pulmonary disease, severe liver disease, concomitant infections
- Patients to use with caution:
- Cardiac disease
- Hypertension
- Elderly
- Severe hypoxemia
- Hyperthyroidism
- Chronic corpulmonale
- Congestive heart failure
- Liver disease
- Peptic ulcer
- Renal impairment
Overdose Effects
Severe cardiac arrhythmias, tonic-clonic seizure, requires discontinuation of treatment, symptomatic treatment of cardiovascular collapse
