Indications
- Treatment of Philadelphia chromosome positive chronic myeloid leukemia in adults
- Treatment of chronic phase and accelerated phase CML in adult patients

Pharmacology
- Inhibition of BCR-ABL tyrosine kinase activity
- Inhibition of TEL-platelet-derived growth factor receptor-beta and FIP1-like-1-PDGFRalpha
- Inhibition of c-Kit receptor kinase
- Potential use for myeloproliferative diseases and gastrointestinal stromal tumours
Dosage & Administration
- Recommended dose for newly diagnosed Ph+ CML-CP: 300 mg orally twice daily
- Recommended dose for resistant or intolerant Ph+ CML-CP and CML-AP: 400 mg orally twice daily
- Administration approximately 12 hours apart
- No food intake for at least 2 hours before and for at least one hour after dose
Interaction
- Reduction in solubility and bioavailability due to drugs inhibiting gastric acid secretion
Contraindications
- Hypokalaemia, hypomagnesaemia, long QT syndrome
- Concomitant use with potent CYP3A4 inhibitors or antiarrhythmics and other QT prolonging drugs
- Concomitant use with potent CYP3A4 inducers
Side Effects
- Rash, pruritus, hepatotoxicity, headache, fever
- GI disturbances, alopecia, fatigue, muscle spasms
- Arthralgia, myalgia, oedema, insomnia, dizziness
- Thrombocytopenia, neutropenia, anaemia, arrhythmias, heart failure
- Elevated AST/ALT, serum lipase, electrolyte imbalances
Pregnancy & Lactation
- Pregnancy Category D
- Use of effective contraception during treatment
- Avoidance of breastfeeding
- No data to support use in pediatrics
- Lower starting dose for patients with hepatic impairment
Precautions & Warnings
- History of pancreatitis, total gastrectomy
- Hepatic impairment, electrolyte imbalances
- Maintenance of hydration and correction of uric acid levels
Overdose Effects
- Symptoms: Neutropenia, vomiting, drowsiness
- Management: Monitor patient and provide supportive treatment
