Categories and Usage
- Indicated for neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Indicated for postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Indicated for fibromyalgia
- Indicated for neuropathic pain associated with spinal cord injury
Pharmacology
- Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Binds with high affinity to the alpha2-delta site in central nervous system tissues
- Mechanism of action not fully elucidated
Dosage
- Maximum recommended dose for DPN: 100 mg three times a day
- Maximum recommended dose for PHN: 75 to 150 mg two times a day or 50 to 100 mg three times a day
- Recommended dose for fibromyalgia: 300 to 450 mg/day
- Recommended dose range for neuropathic pain associated with spinal cord injury: 150 to 600 mg/day
Administration
- Oral route of administration
- Can be taken with or without food
- Pregabalin CR tablet should be administered after an evening meal
- Swallowed whole and should not be split, crushed or chewed
Drug Interaction
- Unlikely to be involved in significant pharmacokinetic drug interactions
Contraindications
- Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components
Side Effects
- Most common side effects in adults: dizziness, somnolence, dry mouth, edema, blurred vision, weight gain
- Most common side effects in pediatric patients for partial onset seizures: increased weight, increased appetite
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Pregnant women should be advised of potential risk to a fetus
- Small amounts detected in the milk of lactating women
- Breastfeeding not recommended due to potential risk of tumorigenicity
Precautions & Warnings
- Angioedema and hypersensitivity reactions may occur
- Increase in risk of suicidal thoughts or behavior
- Respiratory depression may occur when used with concomitant CNS depressants
- May cause dizziness, somnolence, and impair ability to drive or operate machinery
- Increased seizure frequency or adverse reactions may occur if rapidly discontinued
- May cause peripheral edema

- Safety and effectiveness in pediatric patients not established for certain indications
Overdose Effects
- Symptoms include reduced consciousness, depression/anxiety, seizures, and heart block
- No specific antidote available
