Indications

  • Bronchial asthma
  • Bronchospasm
  • Chronic obstructive pulmonary disease (COPD)
  • Pulmonary disease with spastic bronchial component

Pharmacology

  • Selective inhibition of phosphodiesterase 4
  • Relaxation of bronchial smooth muscle
  • Inhibition of platelet activating factor and leukotriene production

Dosage & Administration

  • Elderly dosage: 200 mg tablet two or three times daily
  • Adults dosage: 400 mg tablet two or three times daily
  • Children dosage: 10 ml syrup or 200 mg tablet two or three times daily for >12 years of age, 6-9 mg/kg body weight two times daily for 6-12 years of age
  • If required daily dose of 400 mg, Doxophylline SR tablet to be taken once daily

Interaction

  • Avoid administration with other xanthine preparations
  • Limit consumption of caffeine-containing beverages and food
  • Exercise caution when administering with ephedrine or sympathomimetic drugs
  • Concurrent administration with certain drugs may reduce hepatic clearance

Contraindications

  • Known hypersensitivity to the drug or other xanthine derivatives
  • Acute myocardial infarction
  • Hypotension
  • During lactation

  • Nausea
  • Vomiting
  • Epigastric pain
  • Headache
  • Irritability
  • Insomnia
  • Tachycardia
  • Extrasystoles
  • Tachypnea
  • Hyperglycemia
  • Albuminuria
  • Severe cardiac arrhythmias in case of overdose

Pregnancy & Lactation

  • Caution should be taken during pregnancy and evaluation should be done on a case-by-case basis
  • Contraindicated during lactation

Precautions & Warnings

  • Factors that may reduce hepatic clearance
  • Monitoring of blood levels recommended in certain cases
  • Caution in patients with cardiac disease, hypertension, elderly, severe hypoxemia, hyperthyroidism, liver disease, renal impairment

Overdose Effects

  • Severe cardiac arrhythmias
  • Tonic-clonic seizure
  • Symptomatic treatment of cardiovascular collapse should be instituted
