Indications
- Essential for calcium and phosphate absorption

Pharmacology
- Active form is Calcitriol
- Binds to Vitamin D receptors (VDRs)
- Absorbed in the small intestine
- Metabolized to 25-hydroxy Vitamin D3 and 1,25-dihydroxy Vitamin D3
- Stored in fat and muscle
- Eliminated via faeces and urine

Interaction
- Many drugs can cause Vitamin D3 deficiencies
- Drugs include Magnesium containing Antacids, Digoxin, Thiazide Diuretics, Cholestyramine, Cholistipol, Phenytoin, Phenobarbital, Orlistat, Mineral Oil, Corticosteroids like Prednisolone
Contraindications
- Contraindicated in diseases associated with hypercalcaemia
- Contraindicated in patients with hypersensitivity to Vitamin D3 or its excipients
- Contraindicated in cases of Vitamin D3 toxicity
Side Effects
- Symptoms may include anorexia, lassitude, nausea & vomiting, diarrhea, constipation, weight loss, polyuria, sweating, headache, thirst, vertigo, raised concentrations of calcium and phosphate in plasma and urine
Pregnancy & Lactation
- Safe use up to 4000 IU during pregnancy
- Recommended daily intake for pregnant women is 400 IU
- Excreted in breast milk, overdose in infants from nursing mothers has not been observed
Precautions & Warnings
- Plasma-calcium concentration should be monitored in patients receiving high doses
- Consult a health care practitioner before supplementing with Vitamin D3 if using Digoxin and Thiazide Diuretics, liver or kidney disease, primary hyperthyroidism, lymphoma, tuberculosis, or granulomatous disease
Use in Special Populations
- Safety & efficacy in children under 12 years not established
