Indications
- Breast Cancer treatment

Pharmacology
- Acts by disrupting the microtubular network in cells
- Promotes the assembly of tubulin into stable microtubules
- Inhibits microtubule disassembly
- Binds to free tubulin, decreasing intracellular tubulin concentration
Dosage & Administration
- Administered intravenously every 3 weeks
- Dosage varies for different indications
- Requires premedication with oral corticosteroids
- Adjustable dose
Interaction
- CYP3A4 substrate
- Metabolism may be modified by concomitant administration of certain compounds
Contraindications
- Contraindicated in patients with neutrophil counts <1500 cells/mm³
- Contraindicated in patients with history of severe hypersensitivity reactions to docetaxel or polysorbate 80
Side Effects
- Common adverse reactions include infections, neutropenia, anemia, febrile neutropenia, hypersensitivity, thrombocytopenia, neuropathy, dysgeusia, dyspnea, constipation, anorexia, nail disorders, fluid retention, asthenia, pain, nausea, diarrhea, vomiting, mucositis, alopecia, skin reactions, myalgia
Pregnancy & Lactation
- Can cause fetal harm when administered to a pregnant woman
- No information regarding presence in human milk or effects on breastfed child
Precautions & Warnings
- Administered under supervision of experienced physician
- Higher risk of severe adverse reactions in patients with hepatic impairment
- Should not be given to patients with neutrophil counts <1500 cells/mm³
- Severe hypersensitivity reactions may occur, requiring immediate discontinuation
- Treatment-related acute myeloid leukemia may occur
Overdose Effects
- No known antidote for docetaxel overdose
- Patient should be closely monitored in a specialized unit
- Exacerbation of adverse events may be expected
