Indications
- Symptomatic treatment of allergic conditions of the respiratory tract and skin

Pharmacology
- Phenothiazine derivative
- Blocks postsynaptic mesolimbic dopaminergic receptors
- Strong alpha-adrenergic blocking effect
- Depresses release of hypothalamic and hypophyseal hormones
- Competes with histamine for H1-receptor
- Muscarinic blocking effect
- Reduces stimuli to brainstem reticular system
- Well absorbed from gastrointestinal tract
- Clinical effects appear within 20 minutes after oral administration
- Metabolized by the liver
Dosage & Administration
- Initial dose for adults: one 25 mg tablet at night
- May be increased to two or three 25 mg tablets at night if necessary
- Elderly dosage not specified
- Children treatment with elixir containing 5 mg/5 ml
- Dosage for antihistamine and sedative for children
Interaction
- Enhances the action of anticholinergic agents
- May interfere with immunologic urine pregnancy tests
- Discontinuation required before skin tests using allergen extracts
Contraindications
- Contraindicated for children under two years
- Hypersensitivity to promethazine or excipients
- Should not be used in coma or CNS depression patients
- Avoid in neonates or premature infants
- Avoid in patients using MAO inhibitors within 14 days

Precautions & Warnings
- Avoid in children with signs suggestive of Reye's syndrome
- May thicken or dry lung secretions
- Use caution in patients with asthma, bronchitis, or bronchiectasis
- Care needed in patients with coronary artery disease
- Care needed in patients with narrow angle glaucoma
- Care needed in patients with epilepsy
- Care needed in patients with hepatic insufficiency
