Description
U-Tovas is a selective inhibitor of HMG-CoA reductase, indicated for reducing elevated total cholesterol, LDL cholesterol, apolipoprotein B, and triglycerides levels in various diseases such as familial hypercholesterolaemia, mixed dyslipidemia, hypertriglyceridemia, and more. It is rapidly absorbed after oral administration and extensively metabolized by cytochrome P450 3A4. Atorvastatin is contraindicated during pregnancy and breastfeeding, and should be used with caution in patients with liver disease or renal dysfunction.
DosageAndAdministration
- Adults: Usually 10 mg once daily; may be increased to max. 80 mg once daily
- Child (10-18 years): Initially 10 mg once daily, increased if necessary to max. 20 mg once daily.
ModeOfAction
Atorvastatin is a selective inhibitor of HMG-CoA reductase, lowering plasma cholesterol and lipoprotein levels by inhibiting cholesterol synthesis in the liver and increasing the number of hepatic LDL receptors for enhanced uptake and catabolism of LDL.
Indications
- Reducing total cholesterol and LDL cholesterol in familial hypercholesterolaemia
- Treating patients with dyslipidemia and hypertriglyceridemia
- Preventing cardiac ischaemic events in patients with coronary artery disease
Interaction
- Concurrent administration with cyclosporine, fibric acid derivatives, niacin, erythromycin, or azole antifungals increases the risk of myopathy
- Plasma concentrations may decrease with antacids and increase with erythromycin or oral contraceptives

- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases

- Plasma concentrations are higher in healthy elderly subjects than in young adults
- Renal disease has no influence on the plasma concentrations of U-Tovas
- Hemodialysis is not expected to significantly enhance clearance of U-Tovas
OverdoseEffects
Specific treatment is not available for atorvastatin overdose; the patient should be treated symptomatically and supportive measures instituted, as required.
