Indications
- Relief of symptoms associated with seasonal allergic rhinitis
- Relief of symptoms associated with perennial allergic rhinitis
- Treatment of uncomplicated skin manifestations of chronic idiopathic urticaria
- Treatment of allergen induced asthma
Pharmacology
- Potent H1 receptor antagonist
- No significant anticholinergic effects
- No significant antiserotonic effects
- Inhibits histamine-mediated early phase of allergic reaction
- Reduces migration of inflammatory cells
- Reduces release of mediators associated with late phase of allergic reaction
- Achieves peak plasma concentrations of 257 mcg/L within one hour of administration
- Peak blood levels reached between thirty and sixty minutes after administration of 10 mg dose
- Plasma half-life approximately 11 hours
- Consistent absorption from one subject to the next
- Renal clearance is 30 ml/minute
- Excretion half-life approximately nine hours
- Food does not affect extent of absorption
- Food may slightly reduce the rate of absorption
Dosage & Administration
- Adult dosage: 1 tablet or 2 teaspoonfuls daily
- Children 6 years and older dosage: 1 tablet or 2 teaspoonfuls daily
- Children 2-6 years dosage: 1 teaspoonful once daily or 1/2 teaspoonful twice daily
- Children 6 months to 2 years dosage: 1/2 teaspoonful once daily
- Children 12-23 months dosage: Maximum dose of 1/2 teaspoonful every 12 hours
- Recommended injectable dosage for adults and adolescents: 10 mg administered by intravenous injection
- Recommended injectable dosage for children 6 to 11 years: 5 mg or 10 mg depending on symptom severity administered by intravenous injection
- Recommended injectable dosage for children 6 months to 5 years: 2.5 mg administered by intravenous injection
- Cetirizine injectable dosage form is for intravenous administration only
Interaction
- No clinically significant drug interactions with Theophylline
- No clinically significant drug interactions with Azithromycin
- No clinically significant drug interactions with Pseudoephedrine
- No clinically significant drug interactions with Ketoconazole
- No clinically significant drug interactions with Erythromycin
- No significant drug interactions with other drugs
Contraindications
- Contraindicated in patients with hypersensitivity to Cetirizine
- Contraindicated in patients with hypersensitivity to hydroxyzine
Side Effects
- Most common side effect: somnolence
Pregnancy & Lactation
- US FDA Pregnancy Category: B
- No adequate and well-controlled studies in pregnant women
- Drug excreted in human milk
- Caution should be exercised when administered to a nursing woman
Precautions & Warnings
- Caution should be exercised when driving a car
- Caution should be exercised when operating heavy machinery
Use in Special Populations
- Total body clearance reduced in patients with mild to moderate hepatic and renal impairment
- AUC and half-life increased by about 2 to 3 fold in patients with hepatic and renal impairment
- Plasma levels unaffected by hemodialysis
- Safety demonstrated in pediatric patients aged 6 months to 5 years
- Clearance reduced in elderly patients, proportionate to decrease in creatinine clearance
