Anaya 1 mg (Tablet)

Indications

• Adjuvant treatment of postmenopausal women with hormone receptor-positive early breast cancer
• First-line treatment of postmenopausal women with hormone receptor-positive locally advanced or metastatic breast cancer
• Second-line treatment of advanced breast cancer in postmenopausal women with disease progression following tamoxifen therapy

Pharmacology

• Non-steroidal aromatase inhibitor
• Inhibition of aromatase enzyme to suppress estrogen biosynthesis in peripheral tissues and cancer tissue

Dosage & Administration

• One tablet 1 mg taken once a day
• Continued use for advanced breast cancer until tumor progression
• No recommended dose changes for patients with mild-to-moderate hepatic impairment or renal impairment
• Safety and efficacy not established for pediatric use
• Pharmacokinetics not affected by age

Interaction

• Reduced plasma concentration when co-administered with tamoxifen
• Incompatibility with estrogen-containing therapies
• Unlikelihood of affecting other drugs through cytochrome P450 inhibition

Contraindications

• Contraindicated during pregnancy
• Contraindicated in patients with hypersensitivity to the drug or excipients

Side Effects

• Serious adverse reactions include skin reactions, allergic reactions, and changes in liver function
• Common adverse reactions include hot flashes, asthenia, arthritis, pain, hypertension, depression, nausea, vomiting, rash, osteoporosis, fractures, back pain, insomnia, headache, bone pain, and more

Pregnancy & Lactation

• Pregnancy Category X
• Unknown excretion in human milk
• Recommendation to consider discontinuation of nursing or drug based on importance to the mother

Precautions & Warnings

• Increased incidence of ischemic cardiovascular events in patients with pre-existing ischemic heart disease
• Mean decrease in bone mineral density compared to baseline
• Higher incidence of elevated serum cholesterol compared to tamoxifen

Overdose Effects

• Well-tolerated in clinical trials
• No established single dose resulting in life-threatening symptoms
• Lethality observed in rats at doses greater than recommended human dose
• No specific antidote to overdosage
