Description
- Votrient is indicated for the treatment of advanced renal cell carcinoma (RCC) and soft tissue sarcoma (STS) after chemotherapy.
Dosage and administration
- The recommended starting dose is 800 mg orally once daily without food.
Indications
- Treatment of advanced renal cell carcinoma (RCC)
- Treatment of advanced soft tissue sarcoma (STS) after prior chemotherapy
Pharmacology
- Multi-tyrosine kinase inhibitor
- Inhibitor of VEGFR-1, VEGFR-2, VEGFR-3, PDGFR-α, PDGFR-β, FGFR-1, FGFR-3, Kit, Itk, Lck, c-Fms
- Undergoes metabolism by CYP3A4, CYP1A2, CYP2C8
- Mean half-life of 30.9 hours
- Primarily eliminated via feces
Interaction
- Inhibitors and inducers of CYP3A4 may alter metabolism
- Coadministration with strong CYP3A4 inhibitors increases concentrations
- Avoid concomitant use with strong CYP3A4 inducers
- Substrate of P-glycoprotein (P-gp) and breast cancer resistance protein (BCRP)
- Weak inhibitor of CYP3A4, CYP2C8, and CYP2D6
- Increases incidence of ALT elevations when used with Simvastatin
Contraindications
- Known hypersensitivity to Pazopanib or any other components

- QT prolongation and Torsades de Pointes

- Thromboembolic events
- Gastrointestinal perforation
- Interstitial lung disease/Pneumonitis
- Hypertension
- Hypothyroidism
- Proteinuria
- Tumor lysis syndrome
- Infection
- Increased toxicity with other cancer therapy
Pregnancy and lactation
- Can cause fetal harm
- Advised not to breastfeed during treatment and for 2 weeks after final dose
Precautions and warnings
- Hepatotoxicity observed, especially in patients older than 65 years
- QT interval prolongation risk
- Baseline and periodic monitoring of ECGs and electrolytes recommended
Use in special populations
- Females of reproductive potential advised to use effective contraception
- Male patients advised to use condoms with female partners of reproductive potential
- Safety and effectiveness in pediatric patients not established
Overdose effects
- Dose-limiting toxicity observed at high doses
- No specific antidote for overdosage
- General supportive measures recommended
