Indications
- Acute and chronic psychoses
- Mania
- Schizophrenia
Pharmacology
- Selective monoaminergic antagonist
- High affinity for serotoninergic 5-HT2 and dopaminergic D2 receptors
- Binds to alpha1 adrenergic receptors
- Binds with lower affinity to H1 histamine and alpha2 adrenergic receptors
- No affinity for cholinergic receptors
- Balanced central serotonin and dopamine antagonism
- Reduction of extrapyramidal side effect liability
Dosage & Administration
- Initial dose of 2 mg in 1-2 divided doses on 1st day
- Usual dose range of 4-6 mg daily
- Maximum dose of 16 mg daily
- Different doses for elderly or patients with hepatic or renal impairment
Interaction
- Antagonizes effects of levodopa and dopamine agonists
- Reduces plasma clearance when administered with Carbamazepine
- May decrease the clearance when administered with Clozapine
- Enhances effects of certain antihypertensives
