Indications
- Duodenal ulcer
- Gastric ulcer
- Chronic gastritis
- Prophylaxis of gastrointestinal hemorrhage from stress ulceration
Pharmacology
- Non-systemic
- Minimal absorption from the gastrointestinal tract
- Formation of a chemical complex
- Inhibition of pepsin and bile
Dosage
- Usual dose for adults 2 gm twice daily or 1 gm four times a day
- Maximum daily dose of 8 gm
- Pediatric population safety and efficacy not established
- No special dosage requirements for elderly patients
Administration
To be taken on an empty stomach
Interaction
- Reduction of bioavailability of certain drugs
- Non-systemic interaction in the gastrointestinal tract
- Should not be co-administered with citrate preparations
- Separation of administration from other agents may be necessary for critical bioavailability
- Co-administration with enteral feeds should be separated by one hour to prevent bezoar formation
Contraindications
Contraindicated in patients with hypersensitivity to sucralfate
Side Effects
- Most common adverse events: headache, nausea, abdominal pain, constipation, diarrhea, urticaria
- Bezoar formation in patients with underlying medical conditions or receiving enteral tube feedings
- Episodes of hyperglycemia reported in diabetic patients
Pregnancy & Lactation
Safety not established in pregnant women, caution in breastfeeding women
Precautions & Warnings
- Caution in patients with renal dysfunction
- Not recommended for use in individuals on dialysis
- Use with extreme caution in severe or chronic renal impairment
- Periodic laboratory testing recommended for patients with impaired renal function
- Concomitant use of other aluminium containing medications not recommended
- Bezoars reported in severely ill patients, particularly in those with predisposing conditions or receiving enteral tube feeding
Use in Special Populations
- Not recommended for use in children under 14 years of age
- No dose adjustments necessary for elderly patients
- Use with caution in renal insufficiency patients
- Potential effects on ability to drive and use machines
Overdose Effects
- Most cases remained asymptomatic
- Reported symptoms: abdominal pain, nausea, vomiting
- Minimal risks associated with overdose
