Indications

• Component of first-line therapy
• Metastatic carcinoma of the colon or rectum

Pharmacology

• Derivative of Camptothecin
• Interacts with topoisomerase I
• Prevents religation of DNA single-strand breaks

• Intravenous infusion
• Combination regimens
• Single agent regimens

Interactions

• Diuretics increase risks of dehydration
• Prophylactic dexamethasone enhances lymphocytopenia
• St John's wort and ketoconazole may reduce exposure

• Known hypersensitivity to the drug or its excipients
