Indications
- Treatment of active duodenal ulcer
- Treatment of active benign gastric ulcer
- Symptomatic treatment of erosive or ulcerative gastro-esophageal reflux disease (GERD)
- Long-term management of gastro-esophageal reflux disease (GERD maintenance)
- Symptomatic treatment of moderate to very severe gastro-esophageal reflux disease (symptomatic GERD)
- Treatment of Zollinger-Ellison Syndrome
- In combination with antibacterial therapeutic regimens for the eradication of Helicobacter pylori in patients with peptic ulcer disease
Pharmacology
- Suppresses gastric acid secretion by inhibiting the gastric H+/K+-ATPase
Dosage
- Oral dose for active duodenal ulcer and active benign gastric ulcer is 20 mg once daily in the morning
- Oral dose for erosive or ulcerative GERD is 20 mg once daily for four to eight weeks
- Maintenance dose of rabeprazole sodium 20 mg or 10 mg once daily for long-term management
- Treatment of Zollinger-Ellison Syndrome starting with 60 mg once a day, titrated upwards to 120 mg/day
Administration
Tablets should be taken in the morning before eating and swallowed whole
Interaction
- Potential interaction with compounds whose absorption is pH dependent
- Co-administration with ketoconazole or itraconazole may decrease antifungal plasma levels
- No interaction with liquid antacids observed
Contraindications
- Hypersensitivity to the active substance or excipients
- Contra-indicated in pregnancy and during breastfeeding

Precautions & Warnings
- Symptomatic response does not exclude the presence of gastric or esophageal malignancy
- Regular surveillance for long-term patients
- Possibility of increased risk of hip, wrist, and spine fracture with long-term use, especially in the elderly
- Hypomagnesaemia and vitamin B12 absorption concerns
- Risk of cross-hypersensitivity reactions with other proton pump inhibitors
- Monitoring for blood dyscrasias and hepatic enzyme abnormalities
Use in Special Populations
- No dose adjustment for patients with renal or hepatic impairment
- Not recommended for use in children due to lack of safety and efficacy data
Overdose Effects
- Maximum established exposure up to 60 mg twice daily
- Effects are generally minimal and reversible without further medical intervention
- No specific antidote known, treatment should be symptomatic and supportive
