Description

  • Prestorin is a HMG-CoA reductase inhibitor drug
  • Reduces cholesterol level and triglycerides in the body
  • Useful for conditions like Hyperlipidemia, Hypertriglyceridemia, Type 3 Hyperlipoproteinemia, Stroke

Indications

  • Reducing elevated total-C, LDL-C, ApoB, non HDL-C, TG levels
  • Increasing HDL-C
  • Slowing the progression of atherosclerosis
  • Risk reduction of Myocardial Infarction (MI), stroke and arterial revascularization procedures in patients
  • Treatment strategy to lower total-C and LDL-C
  • Treatment for Pediatric patients 10 to 17 years of age with HeFH after failing an adequate trial of diet therapy

Mode of Action

  • Selective and competitive inhibitor of HMG-CoA reductase
  • Increases the number of hepatic LDL receptors on the cell surface
  • Enhances uptake and catabolism of LDL
  • Inhibits hepatic synthesis of VLDL
  • Reduces the total number of VLDL and LDL particles

Dosage & Administration

  • Usual dose range of 5-20 mg/day
  • Maximum recommended dose is 20 mg/day
  • Recommended starting dose for HomoFH is 20 mg once daily

Interaction

  • Remarkable drug interactions with Cyclosporine, Gemfibrosil, Lopinavir/Ritonavir or atazanavir/ritonavir, Coumarin anticoagulants, Concomitant lipid-lowering therapies

Contraindications

  • Known hypersensitivity to product components
  • Liver disease with unexplained persistent elevations in hepatic transaminase levels
  • Pregnant women and women who may become pregnant
  • Nursing mothers

Side Effects

  • Frequent adverse events related to Prestorin: headache, myalgia, constipation, asthenia, abdominal pain, nausea

Pregnancy & Lactation

  • Administered to women of childbearing age when unlikely to conceive
  • Discontinued immediately if patient becomes pregnant
  • Unknown excretion in human milk

Precautions & Warnings

  • Caution in patients with predisposing factors for myopathy
  • Increased risk of myopathy with concurrent administration of other lipid-lowering therapies
  • Liver enzyme tests to be performed before and at 12 weeks following initiation of therapy

Overdose Effects

  • No specific treatment in the event of overdose
  • Symptomatic treatment and supportive measures instituted as required
  • Hemodialysis does not significantly enhance clearance of Prestorin
