Description

Rosuvastatin is a selective and competitive inhibitor of HMG-CoA reductase, used to reduce cholesterol levels

Indications

  • Treatment of primary hyperlipidemia and mixed dyslipidemia
  • Adjunct to diet to reduce elevated total-C, LDL-C, and TG levels
  • Management of primary dysbetalipoproteinemia (Type III hyperlipoproteinemia)
  • Adjunct to diet for patients with hypertriglyceredemia
  • Slowing the progression of atherosclerosis
  • Risk reduction of myocardial infarction, stroke, and arterial revascularization procedures

Pharmacology

Inhibits HMG-CoA reductase, increases hepatic LDL receptors, and reduces VLDL synthesis

DosageAdministration

  • Heterozygous Familial Hypercholesterolemia in Pediatric Patients:
    • Usual dose range: 5-20 mg/day
    • Maximum recommended dose: 20 mg/day
  • Very High Cholesterol and High Risk of MI or Stroke: 40 mg once daily
  • Homozygous Familial Hypercholesterolemia: 20 mg once daily
  • Dosage in Asian Patients: Initiation with 5 mg once daily
  • Use with Cyclosporine, Lopinavir/Ritonavir or Atazanavir/Ritonavir: Limited dose based on concurrent medication

Interaction

  • Cyclosporine: Increases Prestorin exposure. Dose should be limited to 5 mg once daily.
  • Gemfibrosil: Combination should be avoided. If used together, dose should be limited to 10 mg once daily.
  • Concomitant lipid-lowering therapies: Use may increase the risk of skeletal muscle effects.

  • Known hypersensitivity to product components
  • Liver disease
  • Pregnant women and women who may become pregnant
  • Nursing mothers
