Description
- Neurolin CR is indicated for managing neuropathic pain associated with diabetic peripheral neuropathy (DPN), postherpetic neuralgia (PHN), partial-onset seizures, fibromyalgia, and neuropathic pain associated with spinal cord injury.
- Pregabalin, the active ingredient, is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA) and binds with high affinity to the alpha2-delta site in central nervous system tissues. It exhibits anti-nociceptive and antiseizure effects.
- Dosage varies based on the indication and patient response, ranging from 100 mg three times a day for DPN to 300 to 450 mg/day for fibromyalgia. It should be taken orally, with or without food. Dosing for Pregabalin CR capsule starts at 165 mg once daily and may be increased to 330 mg once daily, with a maximum recommended dose of 660 mg once daily.
- Neurolin CR should be discontinued gradually over a minimum of 1 week to minimize adverse reactions and withdrawal symptoms.
- The most common side effects include dizziness, somnolence, dry mouth, edema, blurred vision, weight gain, and changes in thinking. In pediatric patients, increased weight and appetite are common side effects for the treatment of partial onset seizures.
- Pregabalin is contraindicated in patients with known hypersensitivity to Pregabalin or any of its components, and precautions should be taken in cases of angioedema, respiratory impairment, driving impairment, suicidal thoughts, and peripheral edema.
- Safety and effectiveness in pediatric patients have not been established for certain indications, and caution is advised in cases of pregnancy and lactation.
- In case of overdose, reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, and heart block may occur, and general supportive care including monitoring of vital signs is recommended.
- Neurolin CR is unlikely to be involved in significant pharmacokinetic drug interactions, and it should be stored in a cool & dry place, protected from light & moisture, and out of the reach of children.
