Indications
- Treatment of adult patients with symptomatic anemia associated with chronic kidney disease (CKD)
Description
- Orally administered HIF-PH inhibitor
- Promotes erythropoiesis
- Improves iron regulation
- Overcomes EPO-suppressive effects of inflammation
- Downregulates hepcidin
Pharmacology
- Orally bioavailable
- Hypoxia-inducible factor prolyl hydroxylase inhibitor (HIF-PHI)
- Increases endogenous erythropoietin production
- Reduces hepcidin expression
- Improves iron availability
- Boosts hemoglobin (Hb) levels
- Regulation of genes required for erythropoiesis and iron metabolism
Dosage & Administration
- Oral administration three times per week
- Individualized dose for target Hb levels
- Different starting doses based on patient weight
- Dose adjustment guidelines
- Method of administration: Oral ingestion with or without food
Interaction
- Potential drug-drug interactions with phosphate binders, multivalent cation products, gemfibrozil, probenecid, OATP1B1, BCRP substrates

Precautions & Warnings

- Caution in patients with pre-existing risk factors for TVE
- Caution in patients with a history of seizures
- Avoid in patients with serious signs and symptoms of infection
- Avoid in patients with liver disorder
Use in Special Populations
- Not indicated for use in children
- Not recommended for use in patients with severe hepatic impairment
Overdose Effects
- Increased heart rate
- Likely increase in hemoglobin concentration
- Treatment: Dose reduction or interruption
