Indications
- Treatment of bronchial asthma
- Treatment of bronchospasm
- Treatment of chronic obstructive pulmonary disease (COPD)
- Treatment of pulmonary disease with spastic bronchial component
Pharmacology
- Doxophylline is a novel bronchodilator
- Selective inhibition of phosphodiesterase 4
- Relaxation of bronchial smooth muscle
- Inhibition of platelet activating factor (PAF)
- Decreased affinities toward adenosine A1 and A2 receptors
- Inhibition of leukotriene production
Dosage & Administration
- Elderly dosage: 200 mg tablet two or three times daily
- Adult dosage: 400 mg tablet two or three times daily
- Children dosage (>12 years of age): 10 ml syrup or 200 mg tablet two or three times daily
- Children dosage (6-12 years of age): 6-9 mg/kg body weight two times daily
- Doxophylline SR tablet for daily dose of 400 mg
- Dosage to be as prescribed by the physician
Interaction
- Avoid administering with other xanthine preparations
- Limit consumption of beverages and food containing caffeine
- Exercise caution when administering with ephedrine or other sympathomimetic drugs
- Concurrent administration consideration with erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, propanolol, phenytoin, and other anticonvulsants
- Potential increase in dosage requirement in certain cases

- Known hypersensitivity to the drug or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- Lactation

- Contraindicated during lactation
Precautions & Warnings
- Factors reducing hepatic clearance of xanthine derivatives
- Monitoring of blood levels recommended in certain cases
- Caution in patients with cardiac disease, hypertension, the elderly, severe hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
- Slower clearance in patients with congestive heart failure
- No risk of addiction or dependence
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizure
- Symptomatic treatment of cardiovascular collapse
