Indications
- Progestin for oral contraceptive use in females
Pharmacology
- Drospirenone is a progestin for preventing pregnancy
- Lowers risk of pregnancy by suppressing ovulation
- Spironolactone analogue with anti-mineralocorticoid activity
Dosage & Administration
- Swallow one tablet daily for 28 consecutive days
- Take white active tablet daily for first 24 days and pink inert tablet for next 4 days
- Instructions for starting and switching

- Dose-proportional pharmacokinetics
- Maximum plasma concentrations reached in 2-6 hours
- Concomitant food ingestion has no influence on absorption
Distribution
- 95% to 97% bound to serum albumin
- Apparent volume of distribution approximately 4 L/kg

Contraindications
- Renal impairment
- Adrenal insufficiency
- Progestin sensitive cancers
- Liver tumors or hepatic impairment
- Undiagnosed abnormal uterine bleeding
Side Effects
- Clinically significant Side Effects: Hyperkalemia, bleeding irregularities, and amenorrhea
- Other adverse reactions including acne, headache, nausea, decreased libido
Pregnancy & Lactation
- Little or no increased risk of birth defects during inadvertent use in early pregnancy
- Discontinue if pregnancy occurs
- Negligible amounts excreted in breast milk
- No adverse effects on infants' growth or development
