Indications

Description
- Exerts direct inhibitory effect on the parasympathetic nervous system
- Relaxing effect on smooth musculature
- Direct effect on muscle tissue
- Inhibitory effect on parasympathetic nervous system
Pharmacology
- Non-selective muscarinic acetylcholine receptor antagonist
- Binds with higher affinity to M1 subtype
- Higher affinity for central muscarinic receptors in cerebral cortex
- Lower affinity for peripherally located central muscarinic receptors
Dosage & Administration
- Individualized dosage
- Low initial dose, gradually increased
- Dosing determined by patient's reaction
- Postencephalitic patients may require atropine adjuvant
Interaction
- Additive effects with cannabinoids, barbiturates, opiates, and alcohol
- Increased sedative effects with concurrent use of alcohol or other CNS depressants
- Monoamine oxidase inhibitors and tricyclic antidepressants intensify anticholinergic effects
- Increased risk of tardive dyskinesia during concomitant administration of anticholinergics and neuroleptics
Contraindications
- Hypersensitivity to Trihexyphenidyl
- Narrow angle glaucoma
- Blindness reported due to long-term use in narrow angle glaucoma
