Indications
- Reversal of centrally sedative effects of benzodiazepines
- Termination of general anesthesia induced and maintained with benzodiazepines
- Reversal of benzodiazepine sedation in short diagnostic and therapeutic procedures
- Reversal of paradoxical reactions due to benzodiazepines
- Diagnosis and/or management of benzodiazepine overdose
- Differentiation between involvement of benzodiazepines, other medicines or drugs, or brain damage
- Specific reversal of central effects of benzodiazepines in drug or medicine overdose
Dosage & Administration
- Recommended for intravenous (IV) use only
- Administered by an anaesthesiologist or experienced physician
- Initial dose of 0.2 mg administered IV over 15 seconds
- Second dose of 0.1 mg can be injected if desired consciousness not obtained within 60 seconds
- Usual dose of 0.3-0.6 mg, but individual requirements may vary
- Children >1 year-of-age: Recommended initial dose of 0.01 mg/kg (up to 0.2 mg) administered IV over 15 seconds
Interaction
- Blocks central effects of benzodiazepines by competitive interaction at the receptor level
- Blocks effects of non-benzodiazepine agonists at benzodiazepine receptors
- No pharmacokinetic interaction between ethanol and Anexate
Contraindications
- Known hypersensitivity to the medicine
- Given a benzodiazepine for control of a potentially life-threatening condition
Side Effects
- Well tolerated in adults and children
- Hypersensitivity reactions observed
- Complaints such as feelings of anxiety, palpitations, and fear infrequently observed
- Seizures reported in patients known to suffer from epilepsy or severe hepatic impairment
- Withdrawal symptoms may occur following rapid injection of Anexate
Pregnancy & Lactation
- Safety in human pregnancy not established
- Benefits of medication during pregnancy should be weighed against possible risks to the foetus
- Parenteral administration not contraindicated during lactation
Precautions & Warnings
- Particular caution necessary in cases of mixed-substance overdose
- Not recommended in epileptic patients receiving benzodiazepine treatment for a prolonged period
- Monitoring for resedation, respiratory depression, or other residual benzodiazepine effects necessary
- Use with caution in patients with head injury
- Avoid rapid injection in patients with high dose and/or long-term exposure to benzodiazepines
- Not recommended for treatment of benzodiazepine dependence or for the management of protracted benzodiazepine abstinence syndromes
