Indications
- Therapy for atherosclerotic vascular disease
- Reduction of elevated cholesterol levels
- Treatment for hyperlipidemia
- Adjunct to diet for inadequate cholesterol response
Pharmacology
- Atorvastatin lowers plasma cholesterol
- Ezetimibe inhibits cholesterol absorption by small intestine
- Mechanism of action complementary to statins
Dosage & Administration
- Dosage range of 10/10 mg to 80/20 mg daily
- Starting dose of 10/10 mg or 20/10 mg daily
- Starting dose of 40/10 mg for larger reduction in LDL-C
- Dosage in patients with homozygous familial hypercholesterolemia is 40/10 mg or 80/10 mg daily
Interaction
- Increased risk of myopathy with concurrent administration of certain drugs
- Coadministration with cyclosporine and gemfibrozil should be avoided
- Enhanced risk of skeletal muscle effects when combined with niacin
Contraindications
- Active liver disease
- Hypersensitivity to any component of the preparation

Pregnancy & Lactation
- Pregnancy Category X
- Contraindicated in women who are or may become pregnant
- Discontinuation during pregnancy may have little impact on long-term outcomes
- Unknown excretion in human milk
Precautions & Warnings
- Cases of myopathy reported with atorvastatin and colchicine coadministration
- Temporary withholding or discontinuation in suggestive conditions of myopathy
- Recommended liver enzyme tests prior to initiation
- Caution with drugs decreasing levels of endogenous steroid hormones
Use in Special Populations
- Safety and effectiveness not established in pediatric patients
- No dosage adjustment necessary in geriatric patients
