Indications
- Bronchial asthma
- Bronchospasm
- Chronic obstructive pulmonary disease (COPD)
- Pulmonary disease with spastic bronchial component
Pharmacology
- Novel bronchodilator
- Selective inhibition of phosphodiesterase 4
- Relaxation of bronchial smooth muscle
- Inhibition of platelet activating factor (PAF)
- Inhibition of leukotriene production
Dosage & Administration
- Elderly: 200 mg tablet two or three times daily
- Adults: 400 mg tablet two or three times daily
- Children >12 years of age: 10 ml syrup or 200 mg tablet two or three times daily
- Children 6-12 years of age: 6-9 mg/kg body weight two times daily
- Doxophylline SR tablet to be taken once daily if required daily dose is 400 mg
Interaction
- Avoid administration with other xanthine preparations
- Limit consumption of caffeine-containing beverages and food
- Caution with concurrent administration of ephedrine or sympathomimetic drugs
- Potential interaction with erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, propanolol, phenytoin, and other anticonvulsants

- Known hypersensitivity to doxophylline or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- Lactation

- Severe cardiac arrhythmias

Pregnancy & Lactation
- Use during pregnancy should be evaluated carefully on a case-by-case basis
- Contraindicated during lactation
- Effects on ability to drive and use machines: Does not affect alertness
Precautions & Warnings
- Factors such as age, cardiac disease, liver disease, concomitant infections, and certain drug interactions may affect hepatic clearance
- Caution in patients with cardiac disease, hypertension, hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
- Monitoring of blood levels recommended in cases affecting clearance
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizure
- Discontinuation of treatment may be necessary
- Symptomatic treatment of cardiovascular collapse may be required
