Indications
- Bronchial asthma
- Bronchospasm
- Chronic obstructive pulmonary disease (COPD)
- Pulmonary disease with spastic bronchial component
Pharmacology
Doxofylline selectively inhibits phosphodiesterase 4, relaxes bronchial smooth muscle, inhibits platelet activating factor (PAF) and generation of leukotriene production
Dosage & Administration
- Elderly: 200 mg tablet two or three times daily
- Adults: 400 mg tablet two or three times daily
- Children:
- >12 years of age: 10 ml syrup or 200 mg tablet two or three times daily
- 6-12 years of age: 6-9 mg/kg body weight two times daily
Interaction
Doxofylline should not be administered with other xanthine preparations, caution with ephedrine or other sympathomimetic drugs, caution with concurrent administration of certain drugs
Contraindications
Hypersensitivity to the drug or other xanthine derivatives, acute myocardial infarction, hypotension, during lactation

Pregnancy & Lactation
- Animal tests show no interference with pre and postnatal growth, use during pregnancy to be evaluated carefully on risk-benefit ratio, contraindicated during lactation
Precautions & Warnings
Caution in patients with various conditions such as cardiac disease, hypertension, hypoxemia, hyperthyroidism, liver disease, renal impairment, and those receiving certain concurrent medications
Overdose Effects
Severe cardiac arrhythmias, tonic-clonic seizure, requires discontinuation of treatment, no specific antidote
