Indications

Treatment of major depressive disorder (MDD)

Pharmacology

Enhances central noradrenergic and serotonergic activity
Antagonist at central presynaptic α2-adrenergic inhibitory autoreceptors and heteroreceptors
Moderate peripheral α1-adrenergic antagonist
Moderate antagonist at muscarinic receptors
Potent antagonist of histamine (H1) receptors

Pharmacokinetics

Rapid and well-absorbed after oral administration
Peak plasma levels reached after about 2 hours
Mean half-life of elimination is 20-40 hours
Extensively metabolized and eliminated via urine and faeces

Dosage & Administration

Recommended starting dose of 15 mg/day
Administered in a single dose, preferably in the evening or prior to sleep
Effective dose range of 15 to 45 mg/day
Dose changes should be made at intervals of less than 1 to 2 weeks

Interaction

Clinically significant drug-drug interactions with MAOIs
May interrupt the metabolism or activity of Carbamazepine, Phenytoin or Cimetidine
Should be avoided with Alcohol & Diazepam

Contraindications

Contraindicated in patients with known hypersensitivity to Mirtazapine or its excipients
Concomitant use with MAO inhibitors
