Indications
- Short-term management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Decrease eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid
- Inhibits synthesis of prostaglandins
- Biological activity associated with the S-form
- Highly protein bound
- Metabolized in the liver
- Metabolites and some unchanged drugs excreted in the urine
Pharmacology
- Potent analgesic
- Inhibits the cyclooxygenase enzyme system
- Demonstrates minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours for up to 7 days
- Injection: Single or multiple doses, IV bolus given over no less than 15 seconds, IM administration should be given slowly and deeply
- Onset of analgesic effect within 30 minutes, maximum effect in 1-2 hours
- Duration of analgesic effect is usually 4-6 hours
Interaction
- Other NSAIDs or aspirin increases side effects
- Anti-coagulants enhance anti-coagulant effect
- Beta Blocker reduces anti-hypertensive effect
- ACE Inhibitors increase risk of renal impairment
- Methotrexate enhances toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic before surgery
