Indications

  • Peptic ulcer diseases
  • Gastroesophageal reflux diseases
  • Ulcer induced by non-steroidal anti-inflammatory drugs (NSAIDs)
  • Eradication of Helicobacter pylori (in combination with antibiotics)
  • Zollinger-Ellison Syndrome

Pharmacology

  • Proton pump inhibitor
  • Suppression of final step in gastric acid production
  • Covalent binding to H+/K+ATPase enzyme system
  • Inhibition of both basal and stimulated gastric acid secretion

Dosage & Administration

  • Oral administration
  • Benign gastric ulcer: 40 mg daily in the morning for 4 weeks
  • Duodenal ulcer: 40 mg daily in the morning for 2 weeks
  • Prophylaxis of NSAID-associated gastric or duodenal ulcer: 20 mg daily
  • IV Injection for duodenal ulcer and gastruc ulcer: 40 mg once daily for 7-10 days
  • Long-term management of Zollinger-Ellison Syndrome and other hypersecretory conditions: 80 mg IV every 12 hours

No significant drug interactions observed in clinical studies

Hypersensitivity to any components of the formulation

Side Effects

  • Well tolerated in both short-term and long-term treatment
  • Common: headache, diarrhea
  • Rare: abdominal pain, flatulence, rash, insomnia, hyperglycemia

Pregnancy & Lactation

US FDA Pregnancy Category B. Use during pregnancy only if clearly needed. Excreted in human milk.

Precautions & Warnings

  • Tablet should not be split, chewed, or crushed
  • Long-term therapy may lead to malabsorption of Vitamin B12 or increase risk of osteoporosis related disorders

Overdose Effects

  • No known symptoms of overdosage in humans
  • Not readily dialyzable due to high protein binding
