Pharmacology
- COX-2 specific inhibitor
- Cyclooxygenase-1 (COX-1) responsible for prostaglandin-mediated functions
- COX-2 responsible for pain, inflammation, fever
- Inhibition of COX-1 by nonselective NSAIDs associated with gastric damage
- Dose-dependent inhibition of COX-2 by Etoricoxib
- No inhibition of gastric prostaglandin synthesis

Interaction

Contraindications

- Dry mouth, taste disturbance, mouth ulcer, appetite and weight change
- Atrial fibrillation, transient ischaemic attack, chest pain, flushing, cough
- Dyspnoea, epistaxis, anxiety, impaired mental acuity, paraesthesia
- Electrolyte disturbance, myalgia, arthralgia, confusion, hallucinations
Pregnancy & Lactation
- Not recommended for women attempting to conceive
- Excretion in the milk of lactating rats
- Prohibition of breastfeeding while using Etoricoxib
Precautions & Warnings
- Caution in patients most at risk of gastrointestinal complication with NSAIDs
- Consideration for patients with significant risk factors for cardiovascular events
- Potential effect on renal function and need for monitoring
- Caution in patients with history of cardiac failure, left ventricular dysfunction, hypertension
- Monitoring needed for liver function and discontinuation in case of hepatic insufficiency
- Discontinuation at the first sign of hypersensitivity
- Masking of fever and inflammation, caution with co-administration of warfarin

- Absence of significant toxicity with single and multiple doses
