Indications

• Treatment of hypertension

Pharmacology

• Olmesartan blocks the vasoconstrictor and aldosterone-secreting effects of angiotensin-II
• Hydrochlorothiazide affects the renal tubular mechanisms of electrolyte reabsorption

Dosage & Administration

• Usual starting dose: 20/12.5 mg once daily
• Dose may be titrated at intervals of 2-4 weeks

Interaction

• Olmesartan has no significant drug interactions reported
• Hydrochlorothiazide may interact with alcohol, barbiturates, narcotics, antidiabetic drugs, other antihypertensive drugs, corticosteroids, ACTH, and lithium

Contraindications

• Hypersensitivity to any component of the product
• Anuria
• Hypersensitivity to sulfonamide-derived drugs

• Upper respiratory tract infection
• Urinary tract infection
• Chest pain
• Back pain
• Peripheral edema
• Abdominal pain
• Dyspepsia
• Gastroenteritis
• Diarrhea

Pregnancy & Lactation

• Safety and effectiveness not established in nursing mothers and pregnancy
• Discontinue during these conditions

Precautions & Warnings

• Periodic determination of serum electrolytes
• Hyperuricemia may occur in certain patients receiving thiazide therapy
• Impaired renal function

Use in Special Populations

• Renal Impairment Patients: Usual regimens may be followed if creatinine clearance is >30 ml/min
• Hepatic Impairment Patients: No dosage adjustment necessary
• Paediatric use: Safety and effectiveness not established
• Geriatric use: Dose selection should be cautious

Overdose Effects

• Olmesartan: Manifestation of overdosage would be hypotension and tachycardia
• Hydrochlorothiazide: Signs and symptoms observed are those caused by electrolyte depletion resulting from excessive diuresis
