Indications
- Prophylaxis of gastric and duodenal ulceration in NSAID users at high risk of complications from gastric ulcer
- Healing of established NSAID-induced gastric and duodenal damage
- Healing of gastric and duodenal ulcers in the absence of NSAID therapy
- Induction of labor
- Prevention and treatment of postpartum hemorrhage
Pharmacology
- Quick de-esterification to its free acid in the body
- Maximum plasma concentrations diminished when taken with food
- Antisecretory properties
- Mucosal protective properties
- Increases bicarbonate and mucus production
Dosage & Administration
- 800 mcg daily dosage for benign gastric and duodenal ulceration and NSAID associated ulceration
- 200 mcg 2-4 times daily for prophylaxis of NSAID-induced gastric and duodenal ulcer
- Induction of labor dosage of 25-200 mcg
- Prevention of postpartum hemorrhage dosage of 600 mcg orally immediately following delivery
Interaction
- No clinically significant interaction observed with cardiac, pulmonary, CNS drugs and NSAID's
- Bioavailability is decreased with high doses of antacid
Contraindications
- Contraindicated in individuals with a history of allergy to prostaglandins
- Contraindicated in pregnancy
Side Effects
- Frequent adverse effects involve the GI tract such as diarrhea, abdominal pain, dyspepsia, flatulence, nausea, vomiting, rashes, and dizziness
Pregnancy & Lactation
- Contraindicated in pregnant women
- Use in women of childbearing potential only if on effective contraceptive measures and after negative serum pregnancy test
Precautions & Warnings
- Contraindicated in pregnant women for prevention and treatment of NSAID induced gastric and duodenal ulcer
- Induction of labor contraindicated in specific pregnancy conditions
- Effective contraception method must be used by women of childbearing potential
Use in Special Populations
- Safety and effectiveness not established in children below 18 years
Overdose Effects
- Toxic dose not determined in humans
- Clinical signs may include sedation, tremor, convulsions, dyspnea, abdominal pain, diarrhea, and fever
