Indications

  • Nosocomial pneumonia
  • Community-acquired pneumonia
  • Complicated skin and skin structure infection
  • Uncomplicated skin and skin structure infections
  • Vancomycin-resistant Enterococcus faecium infections

Pharmacology

  • Synthetic antibacterial agent
  • Oxazolidinones class of antibiotics
  • In vitro activity against Gram positive aerobic bacteria
  • Binds to the 23S ribosomal RNA of the 50S subunit

Dosage & Administration

  • Oral route administration
  • IV infusion over a period of 30 to 120 minutes
  • Recommended daily dosage for different infections

Interaction

  • Reversible, nonselective inhibitor of monoamine oxidase
  • Potential interaction with adrenergic and serotonergic agents
  • Advice to avoid foods or beverages with high tyramine content

Contraindications

  • Known hypersensitivity to Linezolid
  • Patients taking medicinal products that inhibit monoamine oxidases
  • Uncontrolled hypertension, pheochromocytoma, thyrotoxicosis
  • Patients taking sympathomimetic agents, vasopressive agents, dopaminergic agents, serotonin re-uptake inhibitors, tricyclic antidepressants, serotonin 5-HT1 receptor agonists, meperidine or buspirone

Side Effects

  • Mostly mild to moderate adverse events
  • Common adverse events: diarrhea, headache, nausea
  • Other adverse events: oral moniliasis, vaginal moniliasis, hypertension, dyspepsia, abdominal pain, pruritus, tongue discoloration

Precautions & Warnings

  • Weekly monitoring of complete blood counts
  • Prompt ophthalmic evaluation for symptoms of visual impairment
  • Not approved for catheter-related bloodstream infections
  • Monitoring for potential increases in blood pressure
  • Immediate medical evaluation for specific symptoms

Overdose Effects

  • No reported cases of overdose
  • Symptomatic and supportive care advised
  • Approximately 30% removed during 3 hours of haemodialysis
