Description
- Nabufen is indicated for relief of signs and symptoms of osteoarthritis and rheumatoid arthritis.
- Nabufen is a non-steroidal anti-inflammatory drug (NSAID) that exhibits anti-inflammatory, analgesic, and antipyretic properties.
- Nabumetone is a prodrug that undergoes hepatic biotransformation to the active component, 6-methoxy-2-naphthylacetic acid (6MNA), a potent inhibitor of prostaglandin synthesis.
- The recommended starting dose for osteoarthritis and rheumatoid arthritis is 1000 mg taken as a single dose with or without food. Some patients may obtain more symptomatic relief from 1500 mg to 2000 mg per day.

- Dosages greater than 2000 mg per day have not been studied.

- Caution should be taken in prescribing Nabumetone to patients with moderate or severe renal insufficiency.
Interaction
- NSAIDs may diminish the antihypertensive effect of ACE-inhibitors.
- This interaction should be given consideration in patients taking NSAIDs concomitantly with ACE-inhibitors.
Contraindications
- Nabumetone is contraindicated in patients with known hypersensitivity to nabumetone or its excipients.
- Nabumetone should not be given to patients who have experienced asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs.
Side effects
- Gastrointestinal symptoms such as diarrhea, dyspepsia, abdominal pain, constipation, flatulence, nausea, vomiting.
- Central Nervous System effects including dizziness, headache, fatigue.
- Dermatologic effects such as pruritus, rash.
- Special Senses effects like tinnitus.
- Miscellaneous effects like edema.
Pregnancy lactation
- Nabumetone is Pregnancy Category C.
- Use during the third trimester of pregnancy is not recommended.
- Nabumetone is not recommended for use in nursing mothers.
Precautions warnings
- NSAIDs have been associated with renal papillary necrosis and other abnormal renal pathology during long-term administration to animals.
- Administration of an NSAID results in a decrease in prostaglandin synthesis and, secondarily, in a reduction of renal blood flow.
Overdose effects
- Symptoms following acute NSAIDs overdoses are usually limited to lethargy, drowsiness, nausea, vomiting, and epigastric pain.
- Gastrointestinal bleeding can occur.
- Hypertension, acute renal failure, respiratory depression, and coma may occur but are rare.
- Symptomatic and supportive care following a NSAID overdose is recommended.
