Pharmacology
- Selective and competitive inhibitor of HMG-CoA reductase
- Enhances uptake and catabolism of LDL
- Inhibits hepatic synthesis of VLDL

Dosage & Administration
- Usual dose range of 5-20 mg/day
- Maximum recommended dose of 20 mg/day
- Recommended starting dose of 20 mg/day for homozygous familial hypercholesterolemia
- Initiation of therapy with 5 mg once daily for Asian patients
- Dose limitation in various drug combination scenarios
- Dosing for patients with severe renal impairment
Interaction
- Increased exposure with Cyclosporine combination
- Increased exposure with Lopinavir/Ritonavir or atazanavir/Ritonavir combination
- Prolonged international normalized ratio (INR) with coumarin anticoagulants combination
- Enhanced risk of skeletal muscle effects with concomitant lipid-lowering therapies

- Increased risk of myopathy with concurrent administration of other lipid-lowering therapies
- Temporary withholding in patients with acute, serious conditions
- Recommended liver enzyme tests before initiation and periodically thereafter
Overdose Effects
- Symptomatic treatment in the event of overdose
- Supportive measures as required
- Hemodialysis does not significantly enhance clearance
