Description
- Zasper is indicated for the relief of chronic heartburn symptoms and other symptoms associated with GERD, healing of erosive esophagitis, maintenance of healing of erosive esophagitis, eradication of Helicobacter pylori infection, Zollinger-Ellison Syndrome, Acid related Dyspepsia, Duodenal & Gastric ulcer.

- Eradication of Helicobacter pylori infection in patients with duodenal ulcer disease
- Treatment of Zollinger-Ellison Syndrome
- Management of Acid related Dyspepsia
- Healing of Duodenal & Gastric ulcer
Pharmacology
- Proton pump inhibitor
- Suppression of gastric acid secretion
- Specific inhibition of H+/K+-ATPase in gastric parietal cell
- Better acid control
- Enteric-coated pellet formulation of esomeprazole magnesium
- Peak plasma levels occur at approximately 1.5 hours after oral administration
- Plasma protein binding is constant over the concentration range of 2-20 mmol/L
- Extensively metabolized in the liver by the cytochrome P450 (CYP) enzyme system
Dosage
- 20 mg or 40 mg Once Daily for Healing of Erosive Esophagitis and Symptomatic GERD
- 10-day Triple Therapy for Helicobacter Pylori eradication
- 20-80 mg once daily for Zollinger-Ellison Syndrome
- 20-40 mg once daily for Acid-related Dyspepsia
- 20 mg once daily for Duodenal ulcer and 20-40 mg once daily for Gastric ulcer
- IV Injection for GERD with Erosive Esophagitis and Risk reduction of rebleeding of Gastric or Duodenal Ulcers
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Instructions for Oral Suspension and IV Injection administration
Interaction
- Metabolized in the liver by CYP2C19 and CYP3A4
- No clinically relevant interactions with specific CYP enzymes expected
- Potential interference with CYP2C19 metabolism
- Possible interference with absorption of certain drugs
- No significant interactions with phenytoin, warfarin, quinidine, clarithromycin or amoxicillin

OverdoseEffects
- Lethal effects observed at high doses in animals
- Major signs of toxicity include reduced motor activity, changes in respiratory frequency, tremor, ataxia, and intermittent clonic convulsions
- No specific antidote known
- Supportive and symptomatic treatment recommended
