Indications

Pharmacology
- Mode of Action: 5HT3 receptor-antagonist
- Blockage of Vagal Afferents: Prevents vomiting reflex
- Central Mechanism: Antagonism of 5HT3 receptors in the central nervous system
- Effect on Chemotherapy-induced Nausea and Vomiting: Antagonism of 5HT3 receptors in both peripheral and central nervous system

Administration
- Steps:
- Tear the pouch carefully along with the edge tear mark
- Put the film on top of the tongue
- Do not chew or swallow the film whole
- Swallow after the film dissolves
- Wash hands after administration
Interaction
Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
Contraindications
- Hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Common Adverse Events:
- Headache
- Constipation
- Diarrhea
- Frequency: Majority mild or moderate in nature
- Chemotherapy-induced Nausea and Vomiting: Approximately 1% rash occurrence
Pregnancy & Lactation
- Carcinogenic Effects: Not observed in animal studies
- Excretion in Breast Milk: Caution advised when administered to nursing women
Precautions & Warnings
- Hypersensitivity Reactions: Reported in patients with hypersensitivity to other 5-HT3 receptor antagonists
- Gastrointestinal Stimulation: Not a peristalsis-stimulating drug
Use in Special Populations
- Renal Function: Dosage recommendation same as general population
- Hepatic Function: Recommendation for severe hepatic impairment
- Pediatric Patients:
- Limited information for patients 4 years of age or younger
- No dosage adjustment needed for patients over the age of 65
