Indications
- Primary hyperlipidemia and mixed dyslipidemia
- Hypertriglyceridemia
- Primary dysbetalipoproteinemia
- Homozygous familial hypercholesterolemia (HoFH)
- Atherosclerosis progression
- Pediatric patients with heterozygous familial hypercholesterolemia (HeFH)
- Risk reduction of Myocardial Infarction (MI), stroke and arterial revascularization procedures
Pharmacology
- Selective and competitive inhibitor of HMG-CoA reductase
- Converts 3-hydroxy-3-methyl glutaryl coenzyme A to mevalonate
- Increases hepatic LDL receptors
- Inhibits hepatic synthesis of VLDL
Dosage & Administration
- Usual dose range of 5-20 mg/day
- Maximum recommended dose of 20 mg/day
- Dose adjustment intervals of 4 weeks or more
- 40 mg once daily for very high cholesterol and high risk of MI or stroke
- Recommended starting dose of 20 mg once daily for homozygous familial hypercholesterolemia
- Consider initiation of therapy with 5 mg once daily for Asian patients
- Limit Rosuvastatin dose to 5 mg once daily with Cyclosporine
- Limit Rosuvastatin dose to 10 mg once daily with Lopinavir/Ritonavir or Atazanavir/Ritonavir
- Consider reduction in Rosuvastatin dosage when used with Niacin or Fenofibrate
- Start dosing at 5 mg once daily for patients with severe renal impairment
Interaction
- Cyclosporine: Increases Rosuvastatin exposure
- Gemfibrosil: Potentiates Rosuvastatin exposure
- Lopinavir/Ritonavir or atazanavir/ritonavir: Increases Rosuvastatin exposure
- Coumarin anticoagulants: Prolongs international normalized ratio (INR)
- Concomitant lipid-lowering therapies: Increases risk of skeletal muscle effects
Contraindications
- Known hypersensitivity to product components
- Liver disease
- Pregnant women and women who may become pregnant
- Nursing mothers
Side Effects

Pregnancy & Lactation
- Administered to women of childbearing age only when conception is highly unlikely
- Discontinue immediately if patient becomes pregnant
- Unknown excretion in human milk
Precautions & Warnings
- Prescribed with caution in patients with predisposing factors for myopathy
- Discontinue therapy if markedly elevated creatinine kinase levels occur
- Temporarily withhold therapy in patients with acute, serious conditions
- Recommended liver enzyme tests before initiation and periodically thereafter
