Description

  • Esuvast is indicated for primary hyperlipidemia, mixed dyslipidemia, hypertriglyceredemia, primary dysbetalipoproteinemia, homozygous familial hypercholesterolemia, slowing the progression of atherosclerosis, and risk reduction of Myocardial Infarction (MI), stroke and arterial revascularization procedures in patients without clinically evident CHD, but with multiple risk factors.

DosageAndAdministration

  • Usual dose range is 5-20 mg/day; 40 mg once daily for very high cholesterol and high risk of MI or stroke; recommended starting dose for homozygous familial hypercholesterolemia is 20 mg once daily; for Asian patients, consider initiation with 5 mg once daily; in specific drug combinations, the dose should be limited to ensure safety.

Pharmacology

  • Rosuvastatin is a selective and competitive inhibitor of HMG-CoA reductase, enhancing hepatic LDL receptor numbers and inhibiting hepatic synthesis of VLDL and LDL particles.

Interaction

  • Remarkable drug interactions include effects with Cyclosporine, Gemfibrosil, Lopinavir/Ritonavir or atazanavir/ritonavir, Coumarin anticoagulants, and concomitant lipid-lowering therapies.

Contraindications

  • Contraindicated in cases of known hypersensitivity to product components, liver disease, pregnancy, and nursing mothers.

SideEffects

  • Frequent adverse events thought to be related to Esuvast include headache, myalgia, constipation, asthenia, abdominal pain, and nausea.

PregnancyAndLactation

  • Should be administered to women of childbearing age only when pregnancy is highly unlikely; therapy should be discontinued immediately if pregnancy occurs.

PrecautionsAndWarnings

  • Should be prescribed with caution in patients with predisposing factors for myopathy; therapy should be discontinued in case of markedly elevated creatinine kinase levels or myopathy.

OverdoseEffects

  • No specific treatment in the event of overdose; patient should be treated symptomatically and supportive measures instituted as required; hemodialysis does not significantly enhance clearance of Esuvast.
