Indications

• Upper respiratory tract infections
• Lower respiratory tract infections
• Genito-urinary tract infections
• Skin and soft tissue infections
• Bone and joint infections
• Other infections

Composition

• 375 mg tablet
• 625 mg tablet
• 1 gm tablet

Pharmacology

• Broad spectrum of bactericidal activity
• Susceptible to degradation by (3-lactamases
• Inactivation of beta-lactamase enzymes
• Optimized absorption at the start of a meal
• Approximately doubles the serum levels with doubled dosage

Dosage

• For adults and children over 12 years
• Different doses for different types of infections
• Reflection of dosage in tablet and suspension form
• IV Injection dosage for adults and children
• Extended treatment period with review

Administration

• Can be taken without regard to meals
• Optimized absorption by taking at the start of a meal
• IV injection suitable for intravenous administration
• Usage within 20 minutes after reconstitution

Interaction

• Prolongation of bleeding time and prothrombin time
• Reduction of efficacy of oral contraceptives
• Increased likelihood of allergic skin reactions with allopurinol

Contraindications

• History of Penicillin hypersensitivity
• Possible cross-sensitivity with other beta-lactam antibiotics
• Contraindicated for patients with previous history of cholestatic jaundice

Side Effects

• Uncommon and mild and transitory in nature
• Reported gastrointestinal side effects
• Rare cases of hepatitis and cholestatic jaundice
• Reported skin reactions and angioedema

Pregnancy & Lactation

• Animal studies showing no teratogenic effect
• Limited use in human pregnancy with no untoward effect
• Trace quantities detected in breast milk during lactation

Precautions & Warnings

• Careful use in patients on anticoagulation therapy
• Adjustment of dosage in case of hepatic or renal impairment
• Maintenance of fluid intake and urinary output during high dose

Use in Special Populations

• Adjustment of dosage in case of renal impairment
• Dose adjustment and regular monitoring for hepatic impairment
• Dialysis impact on serum concentrations and additional dosage

Overdose Effects

• Unlikely occurrence of overdose problems
• Potential gastrointestinal symptoms and electrolyte imbalance
• Removal from circulation by haemodialysis
