Description

Controls epileptic seizures by inhibiting voltage-sensitive sodium channels, stabilizes neuronal membranes and inhibits presynaptic excitatory neurotransmitter release

Mode of Action

  • Inhibits voltage-sensitive sodium channels
  • Stabilizes neuronal membranes
  • Modulates the release of presynaptic excitatory neurotransmitters
  • Binds weakly to various receptors like serotonin 5-HT3, Adenosine A1/A2, dopamine D1/D2, GABA A/B, histamine H1, etc.

Interaction

  • Valproate increases Lamo concentrations by more than 2-fold
  • Carbamazepine, phenytoin, phenobarbital, primidone, and rifampin decrease Lamo concentrations by approximately 40%
  • Estrogen-containing oral contraceptives decrease Lamo concentrations by approximately 50%
  • Protease inhibitors lopinavir/ritonavir and atazanavir/ritonavir decrease Lamo exposure by approximately 50% and 32% respectively

Pregnancy & Lactation

Pregnancy Category C. Lamo is excreted in human milk. A decision should be made whether to discontinue nursing or discontinue the drug, taking into account the importance of the drug to the mother.

Precautions & Warnings

  • Discontinue at the first sign of rash
  • Monitor for blood dyscrasias
  • Monitor for suicidal behavior and ideation
  • Monitor for signs of aseptic meningitis

Use in Special Populations

  • Lamo not demonstrated as safe and effective in very young pediatric patients aged 1 to 24 months for Epilepsy
  • Lamo not established as safe and effective in pediatric patients aged 10 to 17 years for Bipolar Disorder

Overdose Effects

  • Ataxia
  • Nystagmus

  • Decreased level of consciousness
  • Coma
  • Intraventricular conduction delay
