Indications

Description
- Phenyltriazine antiepileptic
- Used to control epileptic seizures
- Inhibits voltage-sensitive sodium channels
- Stabilizes neuronal membranes
- Inhibits presynaptic excitatory neurotransmitter release
Pharmacology
- Inhibition of voltage-sensitive sodium channels
- Modulation of release of excitatory neurotransmitters
- Binding properties to several receptors
- Inhibitory effect on serotonin 5-HT3 receptor
- Binding to multiple adrenergic, dopamine, GABA, histamine, and opioid receptors
- Inhibition of R-type calcium currents
Dosage & Administration
- Escalation regimen for patients older than 12 years with epilepsy

Interaction
- Increase in concentrations with valproate
- Decrease in concentrations with carbamazepine, phenytoin, phenobarbital, primidone, rifampin, and estrogen-containing oral contraceptives
- Decrease in exposure with protease inhibitors lopinavir/ritonavir and atazanavir/ritonavir
Side Effects
- Adult side effects: dizziness, headache, diplopia, nausea, blurred vision
- Children side effects: vomiting, diarrhea, infection, fever, abdominal pain, tremor
Pregnancy & Lactation
- Pregnancy Category C
- Excretion in human milk
- Potential for serious adverse reactions in nursing infants
Precautions & Warnings
- Discontinue at first sign of rash
- Monitor for blood dyscrasias, suicidal behavior, ideation, and aseptic meningitis
