Indications
- Early Menstrual Regulation (MR)
- Termination of pregnancy up to 9 weeks of gestation
Composition
- Each box contains 2 blister strips
- One strip contains 1 tablet of Mifepristone INN 200 mg
- Another strip contains 4 tablets of Misoprostol USP 200 microgram each
Mode of Action
- Mifepristone competes with progesterone at progesterone-receptor sites
- Mifepristone sensitizes the myometrium to the contraction-inducing activity of prostaglandins
- Misoprostol causes myometrial contraction by interacting with specific receptors on myometrial cells
Dosage & Administration
- Mifepristone administration on Day 1
- Misoprostol administration on Day 2
- Post-treatment examination on Day 10 to 14
Interactions
- Potential inhibition of Mifepristone metabolism by Ketoconazole, Itraconazole, Erythromycin, and grapefruit juice
Contraindications
- History of allergy to Mifepristone or Misoprostol
- Confirmed or suspected ectopic pregnancy
- IUD in place
- Chronic adrenal failure
- Haemorrhagic disorders or concurrent anticoagulant therapy
Side Effects
- Nausea, vomiting, and diarrhea with Mifepristone
- Gastro-intestinal side-effects and uterine rupture with Misoprostol
Pregnancy & Lactation
- Mifepristone is indicated for Menstrual Regulation
- Unknown excretion of Mifepristone and Misoprostol through human milk
- Recommendation against Misoprostol administration to nursing mothers
Precautions & Warnings
- Combination of Mifepristone & Misoprostol prescribed for specific condition
- Intra Uterine Device [IUD] removal recommended
- Surgical termination recommended for treatment failures
Use in Special Populations
- Decreased dose of Misoprostol for patients with hepatic disease
Overdose Effects
- Close observation for signs of adrenal failure with Mifepristone overdose
- Supportive therapy for Misoprostol overdose
