Indications
- Adjunctive therapy for primary hypercholesterolemia
- Prevention of cardiovascular events
- Treatment for homozygous familial hypercholesterolaemia (HoFH)
- Use in patients with homozygous familial sitosterolemia
Mode of Action
- Localizes at the brush border of the small intestine
- Inhibits absorption of cholesterol via the sterol transporter, Niemann-Pick C1-Like1 (NPC1L1)
- Results in decreased delivery of cholesterol to the liver
- Reduction of hepatic cholesterol stores
- Increased clearance of cholesterol from the blood
Dosage & Administration
- Recommended dose: 10 mg once daily
- Can be administered with or without food
Interaction
- Potential interaction with fibrates leading to cholelithiasis
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations in serum transaminases
Side Effects
- Generally well tolerated in clinical studies
- Similar incidence of adverse events to placebo
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Use during pregnancy only if the potential benefit justifies the risk to the fetus
