Indications

  • Treatment of acute and chronic psychoses
  • Treatment of schizophrenia
  • Treatment of bipolar disorder
  • Maintenance treatment of bipolar I disorder
  • Prevention of recurrence of manic, depressive or mixed episodes

Pharmacology

  • Belongs to dibenzothiazepine derivatives
  • Antagonist at neurotransmitter receptors
  • Antagonist at serotonin 5HT1A and 5HT2 receptors
  • Antagonist at dopamine D1 and D2 receptors
  • Antagonist at histamine H1 receptors
  • Antagonist at adrenergic alpha1 and alpha2 receptors
  • Mechanism of action is unknown

Precautions & Warnings

  • Risk of suicidal thoughts and clinical worsening
  • Caution in patients with known cardiovascular disease

  • Consideration of dose reduction or discontinuation for tardive dyskinesia
